Abstract
Optical coherence tomography (OCT) is an emerging biomedical imaging modality that uses low-coherence interferometry with broadband light to generate high-resolution, micron-scale cross-sectional images of tissue microstructure. Building upon conventional structural OCT, we have developed advanced imaging methods that incorporate additional contrast mechanisms, including both static and dynamic scattering signals, to enable label-free optical microangiography (OMAG), also known as optical coherence tomography angiography (OCTA). These techniques provide three-dimensional visualization of the microvasculature and blood flow at the capillary level without the need for exogenous contrast agents, opening new opportunities for studying tissue physiology and disease. In this talk, I will introduce the fundamental principles of OCT, describe the key technological advances that enable OCTA, and present representative applications. Examples will include in vivo imaging of cerebral microvascular perfusion in small-animal models and retinal microcirculation in humans, illustrating the broad impact of OCTA in biomedical research and clinical medicine.
