Abstract
Hematopoietic stem cells (HSCs) rely on precisely coordinated metabolic programs to preserve function, adapt to environmental cues, and sustain lifelong hematopoiesis. Mitochondria play a crucial role in supporting diverse metabolic pathways to meet cellular energy demands; however, the mechanisms by which mitochondrial metabolism shapes HSC fate remain elusive. At the Ito Laboratory, we aim to elucidate the regulatory networks governing healthy and malignant HSCs. For example, we have explored how mitochondrial quality control and lipid metabolism shape HSC homeostasis, the roles of extracellular vesicles in maintaining HSC properties, and, more recently, initiated projects targeting mitochondria for therapy in leukemia. In this presentation, I will summarize our progress in understanding how mitochondria and cellular metabolism influence HSC fate. Deeper insights into metabolic regulation will refine HSC transplantation and drive novel therapies for hematologic disorders and malignancies.
