TSLP has been shown to be a key factor in promoting type-2 inflammatory responses in allergy and asthma. We have found a novel role of TSLP in acute type-2 inflammatory responses in the lung-it promotes Th2 differentiation directly in the lung, independent of the draining lymph node, through activation of an interstitial macrophage subset. In addition, the role of TSLP in cancer is just now being appreciated. We have found that TSLP is a critical survival factor for tumor cells in metastatic breast cancer, and it also plays an important role in determining the quality of the tumor microenvironment.
